1. Die Autoren des papers beginnen mit einem Loyalitätsbekenntnis.

2. Sie lügen wie gedruckt.

3. Sie lassen sich vom Animal Health Board (AHB) sponsern (siehe Acknowledgements), welches die Wildtierbekämpfung mit 1080 aktiv unterstützt.

4. Sie schließen mit einer spekulativen Prognose, die 1080-Herstellern kein Klotz am Bein ist.

Übrigens sind seit dieser Prognose 24 Jahre vergangen und Neuseeland ballert mit 1080-Ködern nur so um sich - so viel zur Seriosität und "wissenschaftlichen Objektivität" dieses papers.

Neuseeland hat vor wenigen Jahrzehnten einen äußerst lukrativen Deal mit der fluorchemischen Industrie abgeschlossen. Da gings nur um Bares, nie um Wissenschaft. Ein paar tapfere Einzelgänger versuchen nun, den größtmöglichen Schaden abzuwenden:

Schon 1972 verachteten Fluorchemiker den Einsatz von F-acetat zur Wildtierdezimierung. Die Gründe kannst du folgenden Aussagen...

Nun aber zum Wesentlichen: der chronischen Toxizität.

Demnach wird F-acetat nicht vollständig ausgeschieden.

2) Folgendes ist auch wichtig

"Heidelberger hatte anno 1960 mit seinem Team einen Nachweis von F-acetat im Urin von Versuchstieren und Patienten nach Gabe von 5-FU erbringen wollen, was ihm nicht gelang. Er ging infolgedessen davon aus, dass sich F-acetat als F-citrat im Gewebe anreichert und nicht ausgeschieden wird:
"...it is possible that our failure to demonstrate fluoroacetate in urine samples might indicate that small amounts of fluoroacetate produced metabolically are efficiently converted into tissue fluorocitrate and not excreted."

Diese Annahme wurde von Rudolph Peters (Entdecker der Fluoressigsäre) bestätigt:

"Thus it becomes fluorocitric acid, which is a very potent poison, because minute amounts of it block the normal metabolism of citric acid. It sticks in the tissues."

Wenn es also im Gewebe verbleibt, dann müsste es doch entsprechende Fallberichte über Latenzphasen und chronische Wirkungen geben, oder? Hoppla, was haben wir denn hier...

"Two weeks after ingestion of 500 mg of sodium monofluoroacetate an adult male developed paresthesias and weakness. Nerve conduction velocity studies, an electromyogram, and nerve biopsy suggested polyneuropathy, mainly of the axonal type. Four weeks later the patient exhibited marked motor weakness, muscle atrophy and tenderness, impaired position/vibratory sense, and inability to stand."

Hmm, verzögert einsetzende Neuropathie, Schwäche, Muskelschwund und Körperschwankungen. Klingt sehr flox-kompatibel, würde ich sagen. Interessanterweise handelte es sich hierbei um eine tödliche Dosis (netto 380 mg FA). Das bedeutet, dass nach der "inability to stand" sicherlich noch einige beschissene Dinge mehr passierten, über die uns die Autoren des Fallberichts leider nicht informieren konnten.

Blicken wir doch mal auf die Ebola-Hochburg Sierra Leone, wo F-acetat von einer Pflanze namens Dichapetalum toxicarium (ursprgl. unter dem passenden Begriff ratsbane bekannt) synthetisiert werden kann. Dieses Wissen haben sich verfeindete Stämme und Rattenfeinde zunutze gemacht, weshalb Vergiftungsfälle nicht ausblieben:

"He was given to throw away some fish which had been poisoned by the ground fruit of ratsbane for the purpose of killing the house rats. He from greed ate the largest slice of fish. In about half an hour afterwards he had vomiting followed by looseness of the bowels and general trembling. On his admission into hospital, he again vomited and his condition then as described by himself through an interpreter was as follows: 'He was feeling very weak and unable to walk, his legs were dead, and he was losing power over his arm and he was feeling very bad.' On examination in the ward it was found that the patient was suffering from complete paralysis of the lower extremities. The tendon reflexes were abolished. There was marked hyperaesthesia of the inner side of the thighs and legs. Firm pressure of the muscles of the calves gave unusually severe pain. The action of the bladder and rectum remained undisturbed. Although the pupils were normal, yet the patient's vision was not acute. There was some want of coordinating power in the muscles of the upper extremities. The power of deglutition was not affected. The patient's condition was stationary for about a fortnight, when he began to regain power over the lower limbs. He was able to stand with assistance, but was trembling all over, and on attempting to move he dragged his feet and presented the symptoms of one suffering from ataxia. He remained in hospital slowly improving and was discharged, about two months after admission, not completely well, there still remaining a certain amount of inability to use his limbs."

Fassen wir zusammen: Magen-Darm-Beschwerden, Sehbeschwerden, Schwäche, schmerzende Muskeln, persistierende Lähmung der unteren Extremitäten mit Sehnenbeteiligung.

Next one please, diesmal aus der ZNS-Abteilung:

"Long-term sequelae have been reported. Cerebellar ataxia lasting for 18 months was reported in one patient; memory disturbances and depressive behaviour which resolved were also seen in this patient. Tetraplegia, cogwheel rigidity, grand mal epilepsy, cortical blindness, and divergent strabismus were seen in another patient 9 years after exposure; these features were attributed to cerebral hypoxia at the time of ingestion..."

Dass F-acetat im Säugetier kumuliert, kann man an 3-Fluortyrosin sehen

“The signs of toxicity noted were characterized by periods of convulsion and hyperirritability alternating with intervals of extreme lethargy (Boyer et al., 1941). These were similar to signs described by Litzka (1936a)...“

Ob die Überlebensrate in der Versuchsgruppe mit schwacher Erregerbelastung gegenüber der o. g. Gruppe höher war, lässt sich nicht nachvollziehen, denn hier wurden alle mit gewöhnlichem Ciprofloxacin behandelten Tiere bereits nach 3- bis 7-tägiger Wirkstoffverabreichung getötet (der u. g. Zeitpunkt der Beimpfung mit dem Erreger spielt für die LD50 keine Rolle):
"treatment at 12-h intervals for 3 days... At the end of treatment (7 days after bacterial inoculation), rats were sacrificed...

Jedoch ist für die Bestimmung einer LD50 bei Substanzen, die zu Fluoracetat konvertiert werden können, ein mehrwöchiger Beobachtungszeitraum ausschlaggebend:

3-Fluortyrosin
"Die zuerst versuchte Menge von 30 mg/kg erwies sich bei 5 Tieren angewandt auf die Dauer als nicht tragbar. Nach 2 bis 3 Tagen traten nach der Fütterung Krämpfe auf. In 24 Stunden hatten sich die Tiere erholt, wurden aber immer schlapper und starben nach 9-17 Tagen. Die Substanz kumulierte offenbar..."

"10 mg/kg: 6 Tiere. Die Tiere wurden meist kachektisch, starben nach 4 Wochen oder wurden vorher getötet."

5-Fluorouracil
„The toxicity of both the 5-fluorouracil and the 5-fluoroorotic acid [in mice] is characterized by delayed death, such as has been observed with other carcinostatic compounds. For this reason, a comparatively long observation time of 10-20 days was necessary..."

Fazit: Da alle exponierten Tiere zusätzlich mit einem Erreger beimpft waren und die Autorin eine erhöhte Mortalitätsrate primär auf den Erreger zurückführt bzw. exponierte Ratten mit schwacher Erregerbelastung zu früh tötete, kann die Studie nur bedingt als Referenz für eine ableitbare LD50 dienen. Allerdings stellt sich bzgl. der auffällig niedrigen MTD-Werte und der an Fluoracetatvergiftungen erinnernden Symptomatik einmal mehr die Frage nach der tatsächlichen LD50 der Fluorchinolone.

In diesem Kontext sollte ergänzt werden, dass die MTD für den Fluoracetatvorläufer 5-Fluorouracil (Ratte, i.v.) 50 mg/kg beträgt.

Jedoch hat 5-FU beim Kaninchen eine LD50 von nur 15 (i.v.) bis 19 mg/kg (oral), was in etwa mit den LD50-Werten von 3-Fluortyrosin im Rattenversuch korreliert.

Dies legt nahe, dass die o. g. MTD für 5-FU (Ratte, i.v.) bereits einer LD50 entspricht. Tatsächlich ist die offizielle LD50 für Ratten (oral, i.v.) mit 230 mg/kg ungewöhnlich hoch.

Dieser Wert scheint insofern klärungsbedürftig, als die LD50 des maßgeblichen 5-FU-Metaboliten Fluoracetat beim Kaninchen 0.5 mg/kg beträgt, jedoch bei der Ratte schon ab 0.1 mg/kg beginnt.

Es ist also davon auszugehen, dass auch bei 5-FU an den LD50-Werten geschraubt wurde. Die LD50 der Fluorchinolone dürfte sich in einem Bereich zwischen 30 und 50 mg/kg einpendeln, da FC im Vergleich mit 3-FT und 5-FU eine höhere molare Masse und einen etwas niedrigeren F-Anteil haben.
